    The Semantics of Graph Programs

    GP (for Graph Programs) is a rule-based, nondeterministic programming language for solving graph problems at a high level of abstraction, freeing programmers from handling low-level data structures. The core of GP consists of four constructs: single-step application of a set of conditional graph-transformation rules, sequential composition, branching and iteration. We present a formal semantics for GP in the style of structural operational semantics. A special feature of our semantics is the use of finitely failing programs to define GP's powerful branching and iteration commands

    Mitochondrial Redox Metabolism in Trypanosomatids Is Independent of Tryparedoxin Activity

    Tryparedoxins (TXNs) are oxidoreductases unique to trypanosomatids (including Leishmania and Trypanosoma parasites) that transfer reducing equivalents from trypanothione, the major thiol in these organisms, to sulfur-dependent peroxidases and other dithiol proteins. The existence of a TXN within the mitochondrion of trypanosomatids, capable of driving crucial redox pathways, is considered a requisite for normal parasite metabolism. Here this concept is shown not to apply to Leishmania. First, removal of the Leishmania infantum mitochondrial TXN (LiTXN2) by gene-targeting, had no significant effect on parasite survival, even in the context of an animal infection. Second, evidence is presented that no other TXN is capable of replacing LiTXN2. In fact, although a candidate substitute for LiTXN2 (LiTXN3) was found in the genome of L. infantum, this was shown in biochemical assays to be poorly reduced by trypanothione and to be unable to reduce sulfur-containing peroxidases. Definitive conclusion that LiTXN3 cannot directly reduce proteins located within inner mitochondrial compartments was provided by analysis of its subcellular localization and membrane topology, which revealed that LiTXN3 is a tail-anchored (TA) mitochondrial outer membrane protein presenting, as characteristic of TA proteins, its N-terminal end (containing the redox-active domain) exposed to the cytosol. This manuscript further proposes the separation of trypanosomatid TXN sequences into two classes and this is supported by phylogenetic analysis: i) class I, encoding active TXNs, and ii) class II, coding for TA proteins unlikely to function as TXNs. Trypanosoma possess only two TXNs, one belonging to class I (which is cytosolic) and the other to class II. Thus, as demonstrated for Leishmania, the mitochondrial redox metabolism in Trypanosoma may also be independent of TXN activity. The major implication of these findings is that mitochondrial functions previously thought to depend on the provision of electrons by a TXN enzyme must proceed differently

    Reliability of Synaptic Transmission at the Synapses of Held In Vivo under Acoustic Stimulation

    BACKGROUND:The giant synapses of Held play an important role in high-fidelity auditory processing and provide a model system for synaptic transmission at central synapses. Whether transmission of action potentials can fail at these synapses has been investigated in recent studies. At the endbulbs of Held in the anteroventral cochlear nucleus (AVCN) a consistent picture emerged, whereas at the calyx of Held in the medial nucleus of the trapezoid body (MNTB) results on the reliability of transmission remain inconsistent. In vivo this discrepancy could be due to the difficulty in identifying failures of transmission. METHODS/FINDINGS:We introduce a novel method for detecting unreliable transmission in vivo. Based on the temporal relationship between a cells' waveform and other potentials in the recordings, a statistical test is developed that provides a balanced decision between the presence and the absence of failures. Its performance is quantified using simulated voltage recordings and found to exhibit a high level of accuracy. The method was applied to extracellular recordings from the synapses of Held in vivo. At the calyces of Held failures of transmission were found only rarely. By contrast, at the endbulbs of Held in the AVCN failures were found under spontaneous, excited, and suppressed conditions. In accordance with previous studies, failures occurred most abundantly in the suppressed condition, suggesting a role for inhibition. CONCLUSIONS/SIGNIFICANCE:Under the investigated activity conditions/anesthesia, transmission seems to remain largely unimpeded in the MNTB, whereas in the AVCN the occurrence of failures is related to inhibition and could be the basis/result of computational mechanisms for temporal processing. More generally, our approach provides a formal tool for studying the reliability of transmission with high statistical accuracy under typical in vivo recording conditions

    Global Tipping Points Report 2023: Ch1.2: Cryosphere tipping points.

    Drastic changes in our planet’s frozen landscapes have occurred over recent decades, from Arctic sea ice decline and thawing of permafrost soils to polar amplification, the retreat of glaciers and ice loss from the ice sheets. In this chapter, we assess multiple lines of evidence for tipping points in the cryosphere – encompassing the ice sheets on Greenland and Antarctica, sea ice, mountain glaciers and permafrost – based on recent observations, palaeorecords, numerical modelling and theoretical understanding. With about 1.2°C of global warming compared to pre-industrial levels, we are getting dangerously close to the temperature thresholds of some major tipping points for the ice sheets of Greenland and West Antarctica. Crossing these would lock in unavoidable long-term global sea level rise of up to 10 metres. There is evidence for localised and regional tipping points for glaciers and permafrost and, while evidence for global-scale tipping dynamics in sea ice, glaciers and permafrost is limited, their decline will continue with unabated global warming. Because of the long response times of these systems, some impacts of crossing potential tipping points will unfold over centuries to millennia. However, with the current trajectory of greenhouse gas (GHG) emissions and subsequent anthropogenic climate change, such largely irreversible changes might already have been triggered. These will cause far-reaching impacts for ecosystems and humans alike, threatening the livelihoods of millions of people, and will become more severe the further global warming progresses

    Graph transformation languages are declarative, rule-based languages that abstract from low-level representations of graphs and graph manipulations. Based on formal rewriting approaches, ~aph transformation languages have a strong formal underpinning which facilitates formal reasoning. However, graph transformation languages that are both expressive enough for elegantly solving complex graph problems and also have a complete formal semantics to facilitate reasoning about programs, are surprisingly rare. In this thesis, we introduce the graph programming language GP (for Graph Programs). GP is a practical graph transformation language with a simple syntax and complete formal semantics that facilitates formal reasoning. GP is a further development of the language presented in [HP01]. We introduce the basis of GP in form of conditional rille schemata [PS04]. The language is based on four core constructs whose semantiCs is defined in the style of Plotkin's structural operational semantics [Plo04]. Practicality and suitability for verification is demonstrated on various case studies in the domain of graph algorithms. The language GP is extended with procedures and parameterised calls to form the language GP+.     Microbiota-mediated fine-tuning of the threshold of intestinal inflammasome activation in host-microbial mutualism

    The inflammasome is a complex of proteins that controls the activity of caspase-1, pro-IL-1b and pro-IL-18. It acts in inflammatory processes and in pyropoptosis. The lower intestine is densely populated by a community of commensal bacteria that, under healthy conditions, are beneficial to the host. Some evidence suggests that the gut microbiota influences regulation of the inflammasome. Components of inflammasomes have been shown to have a protective function against development of experimental colitis, dependent on IL-18 production. However the precise mechanisms and the role of the inflammasome in maintaining a healthy host-microbial mutualism remains unknown. To address this question, we have performed axenic (GF) and gnotobiotic in vivo experiments to investigate how the inflammasome components mainly at the level of intestinal epithelial cells (IECs) are regulated under different hygiene conditions. We have established that gene expression of the inflammasome components NLRC4, NLRP3, NLRP6, NLRP12, caspase-1, ASC and IL-18 do not differ between germ-free and colonised conditions under steady-state. In contrast, induction in IL-18 was observed following infection with the pathobiont Segmented Filamentous Bacteria or the pathogen C. rodentium. Additional preliminar findings suggest that a more diverse intestinal flora, like specific pathogen-free (SPF) flora, is more efficient in inducing basal activation of the inflammasome and especially production of IL-18 by IECs, shortly after colonisation. We are also in the process of testing if basal activation of the inflammasome upon intestinal colonization with commensal bacteria helps to protect the host from potential pathobiont bacteria, like C. rodentium, SFB, Prevotella and TM7

    Die Frage, welchen Umfang der Terminus „pflegerischer Nutzen“ umfasst, nimmt in internationalen, wissenschaftlichen, aber auch in gesundheitspolitischen Diskursen eine besondere Rolle ein. Beispielsweise sehen die Rahmenbedingungen für digitale Pflegeanwendungen (DiPA) unter anderem auch den Nachweis eines „pflegerischen Nutzens“ vor. Eine Definition des pflegerischen Nutzens ist sowohl bei DiPA als auch für die Bewertung von Pflegetechnologien nur im Ansatz erfolgt. Vor allem bleibt auch unklar, welche Bedingungen an die Nutzenerfassung für Pflegetechnologien gestellt werden können. Die Arbeitsgruppe Evaluation aus dem „Cluster Zukunft der Pflege“ stellt daher exemplarisch bisherige Evaluationsansätze und Projekterfahrungen in der Nutzenbewertung von Pflegetechnologien aus individueller, organisationaler und gesellschaftlicher Perspektive vor und bietet damit eine Annäherung an ein Bild des pflegerischen Nutzens von Pflegetechnologien, das eine multiperspektivische Nutzenerfassung anhand eines Bottom-up-Ansatzes vorsieht

    Effects of oat β-glucan consumption at breakfast on ad libitum eating, appetite, glycemia, insulinemia and GLP-1 concentrations in healthy subjects

    There is evidence that oat β-glucan lowers appetite and ad libitum eating; however, not all studies are consistent, and the underpinning mechanisms are not entirely understood. We investigated the effects of 4 g high molecular weight (MW) oat β-glucan on ad libitum eating, subjective appetite, glycemia, insulinemia and plasma GLP-1 responses in 33 normal-weight subjects (22 female/11 male, mean age (y): 26.9 ± 1.0, BMI (kg/m ): 23.5 ± 0.4). The study followed a randomised double-blind, cross-over design with subjects fed two test breakfasts with and without oat β-glucan followed by an ad libitum test meal on two different days. Blood samples and ratings for subjective appetite were collected postprandially at regular time intervals. Oat β-glucan increased feelings of fullness (p = 0.048) and satiety (p = 0.034), but did not affect energy and amount eaten at the ad libitum test meal. There was a treatment by time interaction for plasma GLP-1, plasma insulin and blood glucose. GLP-1 was significantly reduced at 90 min (p = 0.021), blood glucose at 30 min (p = 0.008) and plasma insulin at 30 and 60 min (p = 0.002 and 0.017, respectively) following the oat β-glucan breakfast when compared with the control breakfast. Four grams of high MW oat β-glucan lowers appetite but not ad libitum eating and beneficially modulates postprandial glycaemia, it does however, not increase plasma GLP-1 secretion.
